Tristetraprolin对体外血管生成的作用

摘要:

目的:研究tristetraprolin(TTP)在血管生成中的作用.方法:构建可以超表达TTP的重组腺病毒,感染原代内皮细胞后进行体外三维血管生成检测.结果:在体外三维血管生成模型检测中,超表达TTP组从第1天开始就明显地促进血管生成,在之后的几天维持并且加大了这种趋势.进一步的统计分析发现,这种促进作用基本是通过促进血管出芽来实现的,而至少在早期对血管芽的生长基本没有影响.结论:TTP对血管生成的促进作用,更多的是通过促进出芽而不是促进血管芽生长来实现的.在血管生成过程中TTP迅速下调,并维持在一个较低水平上.

Objective To identify the function of tristetraprolin(TTP) in angiogenesis.Method Recombinant adenovirus overexpressing TTP was constructed and was used to infect primary microvascular endothelial cells,which was detected using three-dimensional in vitro angiogenesis assay.Results Overexpression of TTP promoted angiogenesis greatly in vitro.Advanced analysis showed that the promoted function was related to the increased ability of sprouting rather than to sprout growth.Conclusion The down-regulation of TTP plays a promoting role in the process of angiogenesis,via promoting sprouting,rather than vascular sprouting growth.
