磁性纳米四氧化三铁及纳米金协同柔红霉素作用于K562/A02细胞的初步研究

摘要:

目的:研究磁性纳米四氧化三铁(Nano-Fe3O4)及纳米金(Nano-Au)是否具有逆转K562/A02细胞耐药的作用.方法:用MTT法测定盐酸柔红霉素(DNR)对K562/A02和K562细胞的毒性,并观察DNR与Nano-Fe3O4或Nano-Au的联合作用.结果:体积分数为25%的Nano-Fe3O4及Nano-Au能有效增强DNR的细胞毒作用,提高细胞的凋亡率,差异具有显著性.结论:Nano-Fe3O4及Nano-Au结合DNR后均能有效逆转K562/A02细胞的耐药性.

Objective To determine whether the magnetic Nano-Fe3O4 and Nano-Au can reverse the resistance of K562/A02 cells or not.Methods MTT was used to examine the effects of daunomycin with and without the magnetic Nano-Fe3O4 or Nano-Au on K562 and K562/A02 cells.The intracellular free daunomycin concentration and the apoptosis percentage of the K562/A02 cells were measured by flow cytometry and fluorescent microscope.Results Nano-Fe3O4 or Nano-Au at a 25% volumn ratio raised the intracellular daunomycin accumulation of the K562/A02 cells and induced more apoptosis percentage.Conclusion The Nano-Fe3O4 or Nano-Au with daunomycin can reverse the resistance of K562/A02 cells significantly.
