肿瘤热疗用Fe2O3纳米磁性粒子的生物相容性研究

摘要:

目的:研究将要用于肿瘤细胞内热疗的自制纳米级Fe2O3磁性粒子的生物相容性.方法:MTT法评价其体外细胞毒性,溶血试验评价其有无溶血作用,小鼠腹腔注射其无菌生理盐水混悬液以测定其LD50,微核试验评价其有无致畸、致突变作用等.结果:该材料对L-929细胞毒性为0~1级;无溶血作用;昆明小鼠腹腔注射该材料混悬液1-ml,其LD50为5.45-g·kg-1;微核实验结果表明无致畸、致突变作用.结论:就目前所取得的实验结果来看,可认为该材料是一种具有良好生物相容性的材料,对于肿瘤热疗可能具有广阔的应用前景.

Objective  To evaluate the in vitro and in vivo toxicity of selfprepared nanosized F  2O  3 ,which has the potential implication in tumor hyperthermia. Methods  MTT assay was used to evaluate the in vitro cyitoxicity; hemolysis test was carried out to estimate blood toxicity; Fe  2O  3 suspended in sterile 0.9% NaCl were intraperitoneally injected into KM mouse to calculate the LD    50; micronucleus (MN) were reckoned to identify genotoxicity. Results  The experiments showed that the toxicity of the material on mouse fibroblast (L929) cell lines was between 0~1 degree;it had no hemolysis activity; LD    50 arrived at 5.45g·kg  -1 after intraperitoneal injection of 1ml suspension; micronucleus test showed it had no genotoxic effects either. Conclusions  Our results show that the selfprepared nanosized F  2O  3 is a kind of high biocompatibility materials and perhaps is suitable for further application in tumor hyperthermia.
