摘要：

目的:探讨CD3AK细胞在基因治疗中的应用,并对逆转录病毒载体转导CD3AK细胞IL-2基因的方法进行研究.方法:利用逆转录病毒载体PLIL-2SN向人CD3AK细胞中导入IL-2基因.结果:从转导细胞中RT-PCR扩增出347bpNeoR基因特异性片段,并测出培养上清液中IL-2浓度为3.276μg·L-1.结论:转导的IL-2基因得到表达,转导成功.这有助于进一步研究转导后淋巴细胞的功能,以开展基因免疫治疗的研究.

Objective This project was designed to evaluate the potential of CD 3AK cells for immunogene therapy, and discussed the method of retrovirus vector transfer IL 2 gene into CD 3AK cells.Method Retrovirus vector PLIL?2SN bearing IL 2 gene was transferred into human CD 3AK cells.Results 347?bp special Neo R gene was amplified by RT PCR, and 3.276?μg·L -1 of IL 2 was detected in cultural supernatants.Conclusion IL 2 gene transduced CD 3AK cells were obtained, which will be helpful in studying the function of transducted CD 3AK cells and its use in immunogene therapy.
