几种医用材料的细胞生物相容性评价的实验研究

摘要:

目的:通过2种体外细胞毒性试验研究,评价几种生物材料的细胞生物相容性.方法:依据ISO 10993-1.1997及GB/T 16886.1-1997,应用琼脂覆盖法和四甲基偶氮唑盐(MTT)比色法对聚丙烯酰胺水凝胶、壳聚糖和类金刚石等5种生物材料的细胞毒性进行评价.通过建立MTT比色法的L929细胞生长代谢标准曲线,确定了96孔板适宜的细胞接种数.结果:5种生物材料MTT试验中毒性级均为0级,琼脂覆盖法中材料周围及脱色区内细胞溶解评价(Z/L值)均为0/0.结论:5种生物材料具有良好的细胞生物相容性.

Objective  To investigate cytobiocompatibility of some medical materials by two kinds of cytotoxicity tests.Methods  Using agar covering test and 3?(4,5?dimethylthiazole-2-yl)?2,5?diphenyl tetrazolium bromide(MTT) assay,the cytotoxicity of some materials such as polyacrylamide hydrogel,diamond-like carbon(DLC) etc.according to ISO 10993?1.1997 and national standard GB/T?16886.1?1997,was evaluated.During our pre-experiment of MTT assay,a curve of L929 cell growth was drawn to determine the proper planted cell number in each 96 well plate.Results  The toxicity grades of the materials were all zero in MTT assay and the value of Zone/Lysis were 0/0 in agar overlaying assay.Conclusion  All tested materials are proved to have good biocompatibility in vitro.
