C-反应蛋白全身性敲除大鼠模型的繁育及鉴定

摘要:

目的:繁育及鉴定C-反应蛋白(CRP)全身性敲除的大鼠模型。方法:通过TALEN技术构建CRP敲除的杂合子大鼠,并将F1代雌鼠与雄鼠相互交配得到F2代大鼠。通过DNA测序以及ELISA法鉴定CRP敲除大鼠。结果:通过测序发现CRP敲除大鼠的CRP基因1#外显子缺失了TTGGT共5个碱基,ELISA结果显示CRP敲除大鼠中血清CRP含量较野生型相比下调98%。结论:成功构建了CRP全身性敲除大鼠,为后期CRP生物学功能的相关研究提供了动物模型。

Objective:To breed and identify rat C-reactive protein (CRP) knock out models. Methods:The rat CRP knock out heterozygotes models were constructed with TALEN technology, and F2 generation was got by F1 generation copulation. The identification of rat CRP knock out was done by DNA sequencing and ELISA assays. Results:CRP knock out rats had five bases missing in their exon1#. The CRP level of the knock out rats displayed 98% reduction compared with the wild types. Conclusion:We have successfully constructed CRP knock out rat models, which is critical animals for the studies of biological functions of CRP.
