大肠癌组织中存活素、环氧化酶-2、T细胞因子4的表达及三者相关性

摘要:

目的:比较存活素(survivin)、环氧化酶-2(COX-2)、T细胞因子4(TCF-4)在大肠癌中的表达情况,分析三者相关性。方法:利用RT-PCR法检测40例大肠癌患者及癌旁正常组织中survivin、COX-2、TCF-4 mRNA的表达情况;应用免疫组化法检测92例大肠癌和50例癌旁正常组织中survivin、COX-2、TCF-4的表达,比较两种组织中3种蛋白表达差异,并分析三者与大肠癌患者临床病理特征之间的关系;采用Western blotting检测三者在癌旁正常组织和结直肠癌组织中的表达情况。结果:survivin、COX-2、TCF-4蛋白阳性率、蛋白及mRNA表达水平大肠癌组织均高于正常黏膜组织;大肠癌survivin、COX-2、TCF-4的蛋白阳性率均在Dukes分期、淋巴结转移中存在差异(P<0.05),COX-2蛋白表达还在癌组织浸润深度上存在差异(P<0.05)。大肠癌组织COX-2与survivin蛋白阳性表达呈正相关性;TCF-4蛋白阳性表达与COX-2、survivin蛋白阳性表达呈正相关。结论:survivin、COX-2、TCF-4在大肠癌组织中高表达与大肠癌发生、侵袭转移密切相关,联合检测survivin、COX-2和TCF-4有助于大肠癌的早期诊断。

Objective: To compare the expression of survivin, cyclooxygenase-2(COX-2) and T cytokine 4 (TCF-4) in colorectal cancer, and its correlation with pathological features.Methods: RT-PCR method was used to detect the expression of survivin mRNA,COX-2 mRNA and TCF-4 mRNA in 40 patients with colorectal cancer and normal tissues adjacent to cancer. Immunohistochemical method was used to detect the expression of survivin,COX-2,TCF-4 in 92 cases of colorectal carcinoma and 50 cases of normal tissues adjacent to carcinoma. The difference of the expression of three proteins between the two tissues was compared and the relationship between the expression of three proteins and the clinicopathological features of patients with colorectal cancer was analyzed. At the same time, Western blotting was used to detect the expression of these three genes in normal paracancerous tissues and colorectal cancer tissues. Results: The positive rates of survivin, COX-2 and TCF-4, protein level and mRNA expression in colorectal cancer tissues were higher than those in normal mucosa tissues. The expressions of survivin, COX-2 and TCF-4 were all associated with lymphatic metastasis and Dukes staging(P<0.05). COX-2 expression was associated with depth of tumor invasion(P<0.05). There was a positive correlation between the positive expression of COX-2 and survivin protein in colorectal cancer. Positive expression of TCF-4 protein was positively correlated with the positive expression of COX-2 and survivin protein. Conclusion: The high expression of survivin, COX-2 and TCF-4 in colorectal carcinoma is closely related to the occurrence and invasion of colorectal cancer. survivin, MMP-9 and TCF-4 can serve as a parameter for early diagnosis of colorectal carcinoma.
