Objective: To investigate the effects of obesity-related advanced glycosylated end-products on the placenta implantation and embryo implantation ability in pregnant mice. Methods: The adult female BABL/c mice were randomly divided into three groups, 10 mice in each group. The obesity model was established in mice who were fed with high-fat feed. The normal group was fed the same amount of ordinary feed, and the mice were implanted with embryos after the models were made. After 20 days, the mice were anesthetized, sacrificed and collected the blood from eyes, and then a series of indexes were detected, The gene and protein expression were detected by the method of RT-PCR and Western blotting. Results: Compared with the normal group, the weight of mice in the normal group remained basically unchanged, except for the 10W model group (P<0.05) in the 3th,5th and 7th week. The plasma cholesterol, triglyceride, blood glucose, and insulin contents in the 5 W model and the 10 W group were statistically significant compared with the normal group (P<0.05). The result of survival rate of embryos indicated that the live embryos reduced from 93 to 25 in the 10 W group, the survival rate of live embryos in two model groups had statistical significance (P<0.05) compared with the normal group. The RT-PCR results showed that, compared with the normal group, the expression levels of RAGE in the 5 W and 10 W model groups was increasing, while the expression levels of HOXA10 genes showed a decreasing trend. The Western blotting results showed that the expression levels of RAGE protein in the 5 W and 10 W model groups were increasing, the expression levels of HOXA10, P/CAF, FKBP52 and PTEN protein were decreasing. Conclusion: Obesity related advanced glycosylation end product influences the expression of important protein by influencing the RAGE receptor expression, ultimately affects the endothelial function and the ability of embryo and placenta implantation in pregnant mice. |
